Somatic transcription factors as conduits into and out of pluripotency
Abstract
Conversion of somatic cells into pluripotent cells requires precise control of reprogramming transcription factors (TFs). Traditionally, TFs that underpin the somatic state are seen as reprogramming barriers. This project will challenge the current dogma, by revealing how such somatic TFs can cooperate with reprogramming factors to drive transition towards pluripotency in a dosage-dependent manner, and conversely also potentiate exit during differentiation. The resulting framework will provide a knowledge foundation to (1) reconcile paradoxical reports of the function of these TFs in development and disease, and (2) for the establishment of new kinds of commercially relevant reprogramming and guided differentiation strategies.
